Spero Therapeutics Pivots Away From Infectious Disease as Shares Sink to Historic Lows

The market has delivered a brutal verdict on Spero Therapeutics’ corporate reinvention. Shares in the biotech group have tumbled roughly 47 percent over the past month, leaving the stock trading at around EUR 1.01 and hovering dangerously close to its 52-week low. The Relative Strength Index now sits at 23.2 points, a reading that technical analysts typically interpret as deeply oversold territory.

What investors are pricing in is a transformation of the company’s entire identity. Spero is abandoning its roots in infectious disease research to concentrate on immunology and inflammation — a pivot that carries both promise and considerable clinical risk.

A New Pipeline Built on a Single Antibody

The cornerstone of this new direction is an antibody designated SP001, for which Spero holds exclusive development and commercialization rights outside Greater China. The therapy targets IgG4-related disease, a rare chronic inflammatory condition with limited treatment options. Mechanistically, the drug works by blocking CD40L, a protein that plays a central role in numerous inflammatory pathways.

Clinical progress, however, remains distant. A Phase 2 study is not expected to begin until the second quarter of 2027, and the company’s partner, Innovent Biologics, is simultaneously planning a separate trial for Sjögren’s syndrome. With that key milestone many months away, the market is left to judge the company on its financial engineering rather than clinical data.

Royalty Deal Buys Time Without Diluting Shareholders

Unlike many biotechs that turn to equity raises to fund new programs, Spero has opted for a non-dilutive financing route. The company struck a deal with Healthcare Royalty, using future revenue from its FDA-approved antibiotic Utebzi as collateral. GSK handles worldwide commercialization of the drug, with Spero receiving royalties and milestone payments — a portion of which has now been monetized upfront.

That transaction delivered USD 105 million in cash and, according to management guidance, extends the company’s runway into the second half of 2029. The structure spares existing shareholders immediate dilution, though the operational risk of clinical execution remains squarely on the company’s shoulders.

August Report Will Test Investor Confidence

The next major catalyst arrives in August 2026, when Spero publishes quarterly results for the first time since completing its sweeping restructuring. Beyond the usual cash position and study timeline updates, market watchers will be scrutinizing early Utebzi sales figures from GSK — revenue that forms the financial bedrock of the new strategy. Details on trial design and the allocation of capital across the new clinical priorities will also be closely examined.

For a company whose stock has been cut nearly in half within a single month, the report represents more than a routine earnings release. It is the first real opportunity for management to demonstrate that the strategic gamble can translate into tangible progress — and for investors to decide whether the current share price reflects a genuine value opportunity or a warning sign of challenges ahead.
